The A34 will be closed between 20:00 GMT and 06:00 on Monday and Tuesday this week for bridge maintenance near Beedon in West Berkshire.
Narrow lanes will be installed over two nights, on separate carriageways.
There will be two weekend carriageway closures in November, causing “significant delays”, the Highways Agency has warned drivers.
A 68-mile detour will be put in place that will add a minimum of one hour to journey times.
This compares to the 33-mile normal journey via the A34.
College Farm Bridge will be resurfaced and waterproofed and the central reservation barrier reconstructed.
The northbound A34 will be closed on Monday, and the southbound on Tuesday.
The narrow lanes will be in place until 14 November.
During the closures traffic will be diverted via the M4, A404(M), A404 and M40. Non-motorway traffic will be diverted via the A4 and A338.
Local access will be maintained to all interchanges during the southbound closure and a diversion will be in place via Graces Lane and Oxford Road for access to Chieveley and Beedon during the northbound closure.
The works should be completed by the end of November.
